Retatrutide is an experimental, once-weekly triple hormone receptor agonist developed by Eli Lilly that targets GLP-1, GIP, and glucagon receptors to treat obesity and type 2 diabetes. By simultaneously suppressing appetite, slowing digestion, and increasing energy expenditure, it has demonstrated significant weight loss in clinical trials, with participants losing up to 24% of their body weight in Phase 2 studies and an average of 71.2 lbs in a Phase 3 trial for knee osteoarthritis.
The medication is currently in Phase 3 clinical trials, with results expected to conclude in early 2026, potentially leading to FDA approval in 2027. Common side effects are primarily gastrointestinal, including nausea, vomiting, diarrhea, and constipation, which are typically mild to moderate and decrease over time. As it is not yet FDA-approved, it is legally available only to participants in clinical trials, and consumers are warned against purchasing counterfeit or unapproved versions from unauthorized sources.
